In this study we tested classification performance of a sex estimation method from the mandible originally developed by Sella-Tunis et al. (2017) on a heterogeneous Israeli population. Mandibular linear dimensions were measured on 60 CT scans derived from the Czech living population. Classification performance of Israeli discriminant functions (DFs-IL) was analyzed in comparison with calculated Czech discriminant functions (DFs-CZ) while different posterior probability thresholds (currently discussed in the forensic literature) were employed. Our results comprehensively illustrate sensitivity of different discriminant functions to population differences in body size and degree of sexual dimorphism. We demonstrate that the error rate may be biased when presented per posterior probability threshold. DF-IL 1 showed least sensitivity to population origin and fulfilled criteria of sufficient classification performance when applied on the Czech sample with a minimum posterior probability threshold of 0.88 reaching overall accuracy ≥ 95%, zero sex bias, and 80% of classified individuals. The last parameter was higher in DF-CZ 1 which was the main difference between those two DFs suggesting relatively low dependance on population origin. As the use of population-specific methods is often prevented by complicated assessment of population origin, DF-IL 1 is a candidate for a sufficiently robust method that could be reliably applied outside the reference sample, and thus, its classification performance deserves further testing on more population samples.

An increasing number of software tools can be used in forensic anthropology to estimate a biological profile, but further studies in other populations are required for more robust validation. The present study aimed to evaluate the validity of MorphoPASSE software for sex estimation from sexually dimorphic cranial traits recorded on 3D CT models (n = 180) from three populations samples (Czech, French, and Egyptian). Two independent observers performed scoring of 4 cranial traits (2 of them bilateral) in each population sample of 30 males and 30 females. The accuracy of sex estimation using traditional posterior probability threshold (pp = 0.5) ranged from 85.6% to 88.3% and overall classification error from 14.4% to 11.7% for both observers, and corresponds to the previously published values of the method. The MorphoPASSE method is also affected by the subjectivity of the observers, as both observers show agreement in sex assignment in 83.9% of cases, regardless of the accuracy of the estimates. Applying a higher posterior probability threshold (pp 0.95) provided classification accuracy of 97.9% and 93.3% of individuals (for observer A and B respectively), minimizing the risk of error to 2.1% and 6.7%, respectively. However, sex estimation can only be applied to 54% and 66% of individuals, respectively. Our results demonstrate the validity of the MorphoPASSE software for cranial sex estimation outside the reference population. However, the achieved classification success is accompanied by a high risk of errors, the reduction of which is only possible by increasing the posterior probability threshold.

Sex is a biological trait fundamental to the study of hominin fossils. Among the many questions that can be addressed are those related to taxonomy, biological variability, sexual dimorphism, paleoobstetrics, funerary selection, and paleodemography. While new methodologies such as paleogenomics or paleoproteomics can be used to determine sex, they have not been systematically applied to Pleistocene human remains due to their destructive nature. Therefore, we estimated sex from the coxal bone of the newly discovered pelvic remains of the Regourdou 1 Neandertal (Southwest France, MIS 5) based on morphological and metric data employing two methods that have been recently revised and shown to be reliable in multiple studies. Both methods calculate posterior probabilities of the estimate. The right coxal bone of Regourdou 1 was partially reconstructed providing additional traits for sex estimation. These methods were cross validated on 14 sufficiently preserved coxal bones of specimens from the Neandertal lineage. Our results show that the Regourdou 1 individual, whose postcranial skeleton is not robust, is a male, and that previous sex attributions of comparative Neandertal specimens are largely in agreement with those obtained here. Our results encourage additional morphological research of fossil hominins in order to develop a set of methods that are applicable, reliable, and reproducible.

The most significant sexual differences in the human skull are located in the upper third of the face (the frontal bone), which is a useful research object, mainly in combination with virtual anthropology methods. However, the influence of biological relatedness on sexual dimorphism and frontal bone variability remains unknown. This study was directed at sexual difference description and sex classification using the form and shape of the external surface of the frontal bones from a genealogically documented Central European osteological sample (nineteenth to twentieth centuries). The study sample consisted of 47 cranial CT images of the adult members of several branches of one family group over 4 generations. Three-dimensional virtual models of the frontal bones were analyzed using geometric morphometrics and multidimensional statistics. Almost the entire external frontal surface was significantly different between males and females, especially in form. Significant differences were also found between this related sample and an unrelated one. Sex estimation of the biologically related individuals was performed using the classification models developed on a sample of unrelated individuals from the recent Czech population (Cechová et al. in Int J Legal Med 133: 1285 1294, 2019), with a result of 74.46% and 63.83% in form and shape, respectively. Failure of this classifier was caused by the existence of typical traits found in the biologically related sample different from the usual manifestation of sexual dimorphism. This can be explained as due to the increased degree of similarity and the reduction of variability in biologically related individuals. The results show the importance of testing previously published methods on genealogical data.

While season-of-death estimation using cementochronology is routine in archaeozoology, its use is much less frequent in bioarchaeology. Based on the character of the outermost increment (bright or dark), two seasons (spring/summer, autumn/winter) can be distinguished. Although many studies mention its potential and possible use in forensic anthropology or bioarchaeology, few exist with estimation results. This study aimed to apply cementochronology-a histological method based on counting and assessing regular circa-annual acellular cementum increments-to 42 individuals from medieval mass graves from Kutná Hora-Sedlec (Czechia, 14th century) to estimate the season-of-death. The mass graves belong to two stratigraphically distinct groups; written and archaeological sources relate them to two catastrophic events (the famine of 1318 and the plague epidemic of 1348-1350). Using cementochronology, we distinguished two distinct seasons corresponding to the two groups of graves, with individuals from the first group dying predominantly in spring/summer, while those from the second group died in autumn/winter. Taking into account the typical seasonal dynamics of epidemics, the results would be more in line with written sources. However, during the evaluation, we faced difficulties identifying the outermost increment and detecting the dark (thinner) increment; we recommend including only young and middle-aged adults in future studies, due to the difficulty of evaluation, and to consider the readability of the tissue (often affected by diagenesis). In conclusion, cementochronology has potential in the context of estimating the season-of-death, but the technical possibilities for enhancing the outermost increment need to be addressed, and the amount of data analysed expanded.

RESUMO

The article introduces the enhancements made to the IsoArcH database for isotope paleopathology. This includes the addition of new metadata fields, which allow for describing abnormal anatomical or physiological conditions in humans and animals at either the individual or sample level. To showcase the novel features of the database, the article features a unique dataset of carbon and nitrogen isotope values obtained on bulk bone collagen from 42 clinically-documented cases of the Jedlicka pathological-anatomical reference collection, dating from the 19th century CE and curated at the National Museum in Prague, Czechia. The dataset includes 70 combined isotopic measurements from individuals who underwent anatomizations between 1841 and 1900 and had distinct bone diseases/disorders: i.e. syphilis, rickets, osteosarcoma, osteomyelitis, and healed fractures. Finally, the article highlights the value of the data in helping the isotope bioarchaeology and paleopathology communities in their understanding of disease processes.

This work presents an automated data-mining model for age-at-death estimation based on 3D scans of the auricular surface of the pelvic bone. The study is based on a multi-population sample of 688 individuals (males and females) originating from one Asian and five European identified osteological collections. Our method requires no expert knowledge and achieves similar accuracy compared to traditional subjective methods. Apart from data acquisition, the whole procedure of pre-processing, feature extraction and age estimation is fully automated and implemented as a computer program. This program is a part of a freely available web-based software tool called CoxAGE3D. This software tool is available at https://coxage3d.fit.cvut.cz/ Our age-at-death estimation method is suitable for use on individuals with known/unknown population affinity and provides moderate correlation between the estimated age and actual age (Pearson's correlation coefficient is 0.56), and a mean absolute error of 12.4 years.

Proteomics has become an attractive method to study human and animal material, biological profile, and origin as an alternative to DNA analysis. It is limited by DNA amplification in ancient samples and its contamination, high cost, and limited preservation of nuclear DNA. Currently, three approaches are available to estimate sex-osteology, genomics, or proteomics, but little is known about the relative reliability of these methods in applied settings. Proteomics provides a new, seemingly simple, and relatively non-expensive way of sex estimation without the risk of contamination. Proteins can be preserved in hard teeth tissue (enamel) for tens of thousands of years. It uses two sexually distinct forms of the protein amelogenin in tooth enamel detectable by liquid chromatography-mass spectrometry; the protein amelogenin Y isoform is present in enamel dental tissue only in males, while amelogenin isoform X can be found in both sexes. From the point of view of archaeological, anthropological, and forensic research and applications, the reduced destruction of the methods used is essential, as well as the minimum requirements for sample size.

Cementochronology has long been associated in the literature with a high correlation between chronological and estimated age, and low differences between the two ages. The excessive accuracy was rather suspicious, and the method did not even appear in common forensic practice. An important step towards more widespread use of the method is the need to standardize work procedures, including indexes for recording the quality of cementum, preparation of thin sections and the age calculation. In our study, we used the standardized protocol for the preparation of thin sections in a set of Czech modern teeth of known age and sex. In the initial phase, 11.5% of the teeth were discarded due to severe caries in the medial part of the root. In a set of single extractions (55 teeth from 55 individuals), we focused on the detailed results of the age estimation, using precision and accuracy indicators. We also used different dental development data to calculate age, given inconsistencies in the use of eruption / mineralization. In a set of multiple extractions (68 teeth from 22 individuals), intra-individual variability was examined. The result of the application of the standardized protocol is an estimate of age with an absolute inaccuracy of -1.7 years and a relative inaccuracy of 5.4%. Calculation of precision and accuracy in the set of single extractions, however, showed the method's limitations: the imprecision measuring the variability of cementum increments counts increased with chronological age, as did the inaccuracy. The use of different dental development data did not significantly increase the accuracy of the age estimation results. Intra-individual variability remains poorly understood - in the set of multiple extractions the differences within one individual ranged between 0.9 and 10.8 years.

Age-at-death estimation of adult skeletal remains is a key part of biological profile estimation, yet it remains problematic for several reasons. One of them may be the subjective nature of the evaluation of age-related changes, or the fact that the human eye is unable to detect all the relevant surface changes. We have several aims: (1) to validate already existing computer models for age estimation; (2) to propose our own expert system based on computational approaches to eliminate the factor of subjectivity and to use the full potential of surface changes on an articulation area; and (3) to determine what age range the pubic symphysis is useful for age estimation. A sample of 483 3D representations of the pubic symphyseal surfaces from the ossa coxae of adult individuals coming from four European (two from Portugal, one from Switzerland and Greece) and one Asian (Thailand) identified skeletal collections was used. A validation of published algorithms showed very high error in our dataset-the Mean Absolute Error (MAE) ranged from 16.2 and 25.1 years. Two completely new approaches were proposed in this paper: SASS (Simple Automated Symphyseal Surface-based) and AANNESS (Advanced Automated Neural Network-grounded Extended Symphyseal Surface-based), whose MAE values are 11.7 and 10.6 years, respectively. Lastly, it was demonstrated that our models could estimate the age-at-death using the pubic symphysis over the entire adult age range. The proposed models offer objective age estimates with low estimation error (compared to traditional visual methods) and are able to estimate age using the pubic symphysis across the entire adult age range.

This paper summarizes the history of the study of bone surface depressions near the contact surfaces of the sacroiliac joint, or grooves located below the linea terminalis, known in the literature as "scars of parturition" or "pelvic scars". Since the beginning of the 20th century, the "sulcus praeauricularis" has been confused with the "sulcus paraglenoidalis" without a satisfactory explanation of their aetiology. Any groove in the preauricular area of the pelvic bone has been referred to as an indicator of female sex and used in sex estimation in a way that does not respect the nature of sex differences. Novotný (1979) included bone structures, which were described by various authors in both females and males, in his method of holistic evaluation. He proposed a three-step categorization for the evaluation of the bone surface of the pelvic preauricular area (three sub-characteristics scored independently) in order to distinguish the sulcus praeauricularis as a female trait from other structures that may occur in both sexes. However, Novotný's work did not become widely known. Testing of the Novotný method was performed in four groups of individuals of known sex without knowledge of the reproductive history of the females (Novotný 1981; Bruzek 2002; Mikesová 2008) and included a total of 852 pelvic bones (457 male and 395 female). The results showed that female morphology, which is characterized by the presence of various forms of sulcus praeauricularis, occurred in a total of 270 of 395 females (68.3%). Female morphology was found in only 5 of 457 males (1.1%). Male morphology was observed in 419 of 457 males (91.7%), but also in 11.1% of females (44 of 395). Indeterminate or ambiguous morphology was found in 33 of 457 males (7.2%) and 81 of 395 females (20.5%). The application of the Novotný method showed that of the total 275 specimens with female morphology, 98.2% were females and only 1.8% were males. It is therefore appropriate to use this reliable method to estimate the sex of skeletal samples. To test the relationship between morphology and obstetric history in females it is necessary to use a sample with known parity.

ABSTRACT: The examination of documented skeletal remains provides an exceptional opportunity for biohistorical research to answer questions about an individual's life and death. Research in this area also makes it possible to assess the reliability of historical records from the period of interest, which is often the subject of discussion, especially in cases of historically known individuals. The remains of K.B.C. (1895-1940), a prominent local landowner and politician, were exhumed because of the repair of a family tomb in Jíloviste, Czech Republic. The aim of this study was to analyze pathological changes in his bones and to interpret these by comparing them with the results of a historical medical records review of private family and public archives regarding his diseases and death, thus verifying their credibility. Morphological and X-ray examinations of the bones revealed several serious pathological changes, whose presence fully corresponded to the studied documents. This showed the records' reliability, and it was thus possible to accurately interpret the lesions found. The results demonstrated the need for interdisciplinary collaboration in the analysis of such cases, including the assistance of the living descendants of the studied individuals, if possible.

The aims of this study are first, to test the relevance of the three non-metric traits in the preauricular region established by Bruzek (2002) for identifying the female population, and second, to test the relationship between the morphology of the preauricular area and parity. Age and pelvic dimensions are also analysed as potential contributing factors. The group under study, derived from the SIMON identified skeletal collection (Vaud, Switzerland), comprises individuals living at the end of the 19th and in the first half of the 20th centuries. The sample consists of 504 coxal bones (including 236 pairs) from 268 individuals (133 females and 135 males), aged 16 to 81, with age classes equally represented. The obstetric history is known for a subgroup of 99 females. Using the three visual criteria employed together, the results of sex estimation indicate that both the indeterminate rate (11.3% for observer 1 and 7.7% for observer 2) and the classification error rate (2.2% and 9.2%, respectively) are low. The study then shows that the preauricular groove, as defined by Bruzek (2002), is present in significant proportions in both nulliparous females (73.8%) and females who have given birth (79.8%), and that therefore it cannot be considered an indicator of obstetric history. The number of children does not seem to be an explanatory factor, but age contributes to the shaping of these bony imprints. The preauricular groove is already recognizable in close to 60% of females aged 16 to 39 years, but the bony changes are more marked with increasing age, leading to a decrease in both sex diagnostic error and indeterminacy. Correlation between a hyperfeminine morphology of the pelvis and a highly feminine development of the preauricular area may indicate a biomechanical contribution.

OBJECTIVE: To determine the degree of similarity of biologically related individuals according to the occurrence of skeletal developmental anomalies (SDA), to see whether these anomalies reflect documented biological relationships. MATERIAL AND METHODS: The sample consists of the skeletal remains of seven members of the noble Swéerts-Sporck family from the 17th-20th centuries. Eighty-nine SDA were examined using morphological assessment, X-ray and CT. The degree of similarity was calculated using a similarity coefficient (Cvrcek et al., 2018). RESULTS: There were three shared SDA in the sample (cranial shift at the C-T border, cervical ribs, hypoplasia of rib 12), and another fifteen individual SDA were reported. The degree of similarity between individuals supports their documented relationships. The greatest similarity was found in closely related individuals such as father/son or siblings, and the least between unrelated individuals. CONCLUSIONS: SDA can be used as a supportive tool for detecting family relationships. The results correspond to the conclusions of earlier analyses of non-metric traits and frontal sinuses in the same sample: the smaller the biological distance between individuals, the greater the degree of their similarity. SIGNIFICANCE: Using unique human skeletal collections, this communication contributes to the expansion of knowledge about the familial occurrence of SDA. LIMITATIONS: The small number of individuals limits the use of statistical approaches. SUGGESTIONS FOR FURTHER RESEARCH: The results call for research on this topic using a larger sample with known genealogical data and the same approaches, to confirm our conclusions.

Skeletal developmental anomalies (SDA) are a subject of constant interest across scientific disciplines, but still mostly as isolates and curiosities. The aim of this study was to find out to what extent the occurrence of SDA reflects documented biological relationships. The skeletal remains of 34 individuals with known genealogical data were available, members of one family over four generations (19th to 20th centuries, Bohemia, Czech Republic), including some inbred individuals. The occurrence of 89 SDA was assessed on the basis of scopic morphological evaluation and X-ray and CT examinations. The degree of similarity between individuals was calculated using a "similarity coefficient" (SC). A linear model was used to test the relationship between positive values of the SC and the relatedness of biologically related individuals. Simultaneously, based on population frequencies of the evaluated anomalies, those that could be considered familial were recorded. A statistically significant relationship between morphological similarity and the biological distance between individuals was found. The greatest similarity was found among close relatives such as parents and children, siblings, or grandparents and grandchildren. The effect of increased consanguinity on the occurrence of anomalies was not confirmed, however. Seventeen SDA shared by closely related individuals were found in the sample, supporting the documented family relationships among them. Eleven of these were selected as possibly familial, but only five were statistically significant: an elongated styloid process, a cervical block vertebrae (arch, facet joints), hamate hamulus aplasia, anteater nose sign, and incomplete fusion of the S1 spinous process. There were also 28 cases of individual occurrences of 17 different SDA, without connection to the documented relationships between individuals.

OBJECTIVES: The appearance of sexually dimorphic traits varies depending on the type of bone, age, environmental and genetic factors and is closely linked to skeletal maturation sequence. Subadult sex estimation currently shows inconsistent accuracy and methods do not incorporate indicators of maturation. The goal of this study is to apply the Santos et al. (2019) adult sex estimation method on virtually reconstructed subadult os coxae and account for pelvic maturation. MATERIAL AND METHODS: The right os coxae of 194 female and male individuals aged 11-30 years from Marseille, France were virtually reconstructed from computed tomography (CT) scans. Santos et al.'s (2019) 11 traits were scored as female, male, or indeterminate. Maturation of 10 pelvic epiphyseal sites was scored using a four-stage system (0-3) to obtain a composite maturity score from 1 to 30. RESULTS: Three maturity groups were identified based on composite maturity scores ranging from 0 to 30. Individuals with a composite maturity score of 15 or higher showed 98 % sex estimation accuracy and a 6 % indeterminate rate. Scores of 2 for the ischiatic tuberosity or 1 for the anterior superior iliac spine can be used as proxies for a composite maturity score of 15 and application on incomplete bones. DISCUSSION: Sexual dimorphism was observed in the epiphyseal maturation sequence and the development of sexually dimorphic pelvic traits. The Santos et al. (2019) method is applicable on immature individuals who meet a maturation threshold with comparable accuracy to adults, without relying on known or estimated age.

Modern humans expanded into Eurasia more than 40,000 years ago following their dispersal out of Africa. These Eurasians carried ~2-3% Neanderthal ancestry in their genomes, originating from admixture with Neanderthals that took place sometime between 50,000 and 60,000 years ago, probably in the Middle East. In Europe, the modern human expansion preceded the disappearance of Neanderthals from the fossil record by 3,000-5,000 years. The genetic makeup of the first Europeans who colonized the continent more than 40,000 years ago remains poorly understood since few specimens have been studied. Here, we analyse a genome generated from the skull of a female individual from Zlatý kun, Czechia. We found that she belonged to a population that appears to have contributed genetically neither to later Europeans nor to Asians. Her genome carries ~3% Neanderthal ancestry, similar to those of other Upper Palaeolithic hunter-gatherers. However, the lengths of the Neanderthal segments are longer than those observed in the currently oldest modern human genome of the ~45,000-year-old Ust'-Ishim individual from Siberia, suggesting that this individual from Zlatý kun is one of the earliest Eurasian inhabitants following the expansion out of Africa.

Biomechanical load and hormonal levels tended to change just like the soft and skeletal tissue of the elderly with age. Although aging in both sexes shared common traits, it was assumed that there would be a reduction of sexual dimorphism in aged individuals. The main goals of this study were (1) to evaluate age-related differences in cranial sexual dimorphism during senescence, (2) to determine age-related differences in female and male skulls separately, and (3) to compare skull senescence in Czech and French adult samples as discussed by Musilová et al. (Forensic Sci Int 269:70-77, 2016). The cranial surface was analyzed using coherent point drift-dense correspondence analysis. The study sample consisted of 245 CT scans of heads from recent Czech (83 males and 59 females) and French (52 males and 51 females) individuals. Virtual scans in the age range from 18 to 92 years were analyzed using geometric morphometrics. The cranial form was significantly greater in males in all age categories. After size normalization, sexual dimorphism of the frontal, occipital, and zygomatic regions tended to diminish in the elderly. Its development during aging was caused by morphological changes in both female and male skulls but secular changes must also be taken into account. The most notable aging changes were the widening of the neurocranium and the retrusion of the face, including the forehead, especially after the age of 60 in both sexes. Sexual dimorphism was similar between the Czech and French samples but its age-related differences were partially different because of the population specificity. Cranial senescence was found to degrade the accuracy of sex classification (92-94%) in the range of 2-3%.

The skull, along with the pelvic bone, serves an important source of clues as to the sex of human skeletal remains. The frontal bone is one of the most significant sexually dimorphic structures employed in anthropological research, especially when studied by methods of virtual anthropology. For this reason, many new methods have been developed, but their utility for other populations remains to be verified. In the present study, we tested one such approach-the landmark-free method of Bulut et al. (2016) for quantifying sexually dimorphic differences in the shape of the frontal bone, developed using a sample of the Turkish population. Our study builds upon this methodology and tests its utility for the Czech population. We evaluated the shape of the male and female frontal bone using 3D morphometrics, comparing virtual models of frontal bones and corresponding software-generated spheres. To do so, we calculated the relative size of the frontal bone area deviating from the fitted sphere by less than 1 mm and used these data to estimate the sex of individuals. Using our sample of the Czech population, the method estimated the sex correctly in 72.8% of individuals. This success rate is about 5% lower than that achieved with the Turkish sample. This method is therefore not very suitable for estimating the sex of Czech individuals, especially considering the significantly greater success rates of other approaches.

The evaluation of frontal sinus similarity is one way to detect biological relationships, especially in small groups, including families of historically known personalities. However, possibilities for studying this issue are currently limited. This contribution deals with the frontal sinuses of a rare osteological sample with known genealogical data, members of the noble Swéerts-Sporck family from the 17th to 20th centuries. The aim is to verify whether the frontal sinuses reflect documented family relationships. Basic dimensions of the frontal sinus such as total surface area and volume, and maximum height and width, and also morphology and anatomical features were evaluated using computed tomography scans. The portions of the frontal sinus above the "external supraorbital line" were analyzed. The degree of similarity between biologically related individuals was determined for each variable and compared with their known biological distance. The degree of similarity based on dimensions was evaluated using both the unadjusted measured data and standardized data adjusted to size. For the unadjusted dimensions, a positive correlation between morphological similarity and biological relatedness was apparent. On the other hand, no positive correlation was apparent for most of the standardized data. Only total volume showed a very weak indication of a positive trend in the standardized data, but this was weaker than in the original values. A positive quantifiable relationship between morphological patterns and biological distance is not clearly indicated. However, nonmetric features do support the documented relationships of the individuals.
